Reproducibility in LC-MS analysis of complex peptide mixtures generated from cellular lysates

Abstract

The evaluation of reproducibility in analysis of complex peptide mixtures generated from whole cell lysates was discussed. LC-MS on an electrospray ion trap mass spectrometer (ITMS) and an electrospray Fourier-transform mass spectrometer (FT-MS) were used for the generation of complex peptide mixtures from whole cell lysates. The analysis of whole cell lysates by LC-MS/MS on an ITMS and LC-MS on an FT-MS were used for more accurate analysis of proteomes. The experiment consisted of five replicate injections by an autosampler on to a C18 nanocolumn system connected to a Finnigan nanospray source on the ITMS.
