Abstract
Integration of top-down and bottom-up mass spectroscopy (MS) method for detection of proteolytic peptides from a complex protein sample was discussed. Bacterial and yeast cells were cultured to suitable levels and a soluble protein fraction was prepared by centrifuging and separating the non-soluble material. The bottom-up MS was accomplished with a Thermo Finnigan LCQ instrument while the top-down MS was conducted with a IonSpec 9.4 FTICR-MS instrument. Results show that the approach provides the most comprehensive coverage of not only protein identification and cataloging, but also the exact molecular form of the protein on the cell.
