Integration of evanescent excitation (EE)-based chip with microfluidic channels for upright and inverted microscope observations

Abstract

We present the integration of a previously reported EE-based chip with both inverted and upright fluorescent microscopes for more demanding applications. Two types of sample delivery platforms, PDMS and glass microchannels, are easily integrated depending on the magnification of an objective lens needed to visualize a target. Although any configuration is achievable regardless of microscope, objective lens, and channel type, here we performed two experiments to demonstrate versatility of the EEbased chip: One is the velocity field measurement of nanobeads in a PDMS microchannel. The other is λ-DNA detection in a thin glass microchannel. Our miniaturized chip could provide a low-cost access to total internal reflection fluorescent microscopy (TIRFM) on both conventional upright and inverted microscopes for μ-TAS applications.
